ABSTRACT

OBJECTIVE@#In order to select the proper size of prosthesis ball head, the biomechanical changes of hip joint using different diameter prosthesis ball head was studied by three-dimensional finite element analysis.@*METHODS@#The thin-layer CT data and related parameters of artificial femoral head prosthesis were used to establish the finite element model of hip joint after artificial femoral head replacement with different ball diameter of prosthesis(M0:preoperative model;M1:ball head diameter=original femoral head diameter;M2:ball head diameter=original femoral head diameter+1 mm;M3:Ball head diameter= Original femoral head diameter -1 mm;M4:ball head diameter =original femoral head diameter -2 mm). Loading the joint forces and related muscle loads were loaded, and the stress distribution and change of bone and cartilage around acetabulum were analyzed by simulating the standing state of one foot when walking slowly.@*RESULTS@#(1) In M1 to M4, the stress concentration in pelvis was different. The peak value of Von Mises stress in the pelvis of M3 was 44.8 MPa, which was the closest to that before operation, with an increment of 13.4%. The displacement of the pelvis of M3 was the smallest in the four groups after operation, with an increment of 1.40 mm. The next was M1, with a peak value of 47.3 MPa, with an increment of 19.7%, and a pelvic displacement of 1.59 mm. (2) In acetabulum area, the peak value of Von Mises stress in M3 was 23.3 MPa, which was the closest to that before operation, with an increment of about 6.3%, followed by M1, with a peak value of 24.0 MPa and an increment of about 8.1%. (3) On the acetabulum cartilage, the stress distribution of M1 and M3 was similar to that before operation, and the peak value of Von Mises stress of M3 was 18.5 MPa, which was the closest to that before operation, followed by M1, which was 22.5 MPa. (4) M1 to M4 showed different degree of stress concentration in the outer upper quadrant of the artificial femoral head, but showed stress shielding under it;among them, the von Mises stress distribution of M3 was more uniform than that of other models, and its peak value (70.8 MPa) was the lowest in each group, followed by M1 (80.7 MPa).@*CONCLUSION@#When femoral head replacement is performed, it is suggested that the prosthesis ball head with the diameter less than 1 mm and the prosthesis ball head with the same diameter as the original femoral head should be used first, so as to obtain the closest natural mechanical characteristics of the hip joint before the replacement and reduce the risk of complications caused by size differences.

ABSTRACT

AIM To establish an HPLC-DAD method for the simultaneous content determination of six constituents in Maiwei Dihuang Pills (Ophiopogonis Radix,Schisandrae chinensis Fructus,Rehmanniae Radix Praeparata,etc.).METHODS The analysis of 50% methanol extract of this drug was performed on a 35 ℃ thermostatic Agilent ZORBAX SB-C18column (4.6 mm × 150 mm,5 μm),with the mobile phase comprising of acetonitrile-water (containing 0.2% phosphate acid) flowing at 0.8 mL/min in a gradient elution manner,and the detection wavelengths were set at 220,230,236 and 274 nm.RESULTS Deoxyschizandrin,schizandrin B,schisandrin,paeoniflorin,paeonol and loganin showed good linear relationships within the ranges of 10-70,6.5-45.5,33.5-234.5,17-119,31-217 and 34-238 μg/mL (r >0.990 0),whose average recoveries (RSDs) were 99.6% (1.7%),100.4% (1.8%),100.7% (1.8%),102.9% (1.7%),102.2% (1.5%) and 99.7% (1.2%),respectively.CONCLUSION This simple and reproducible method can be used for the rapid quality control of Maiwei Dihuang Pills.

ABSTRACT

PURPOSE: Numerous studies have assessed the association of SP110 gene variants with tuberculosis (TB), but the results were inconsistent. Through a comprehensive review and meta-analysis, our study aimed to clarify the nature of genetic risks contributed by 11 polymorphisms for the development of TB. MATERIALS AND METHODS: Through searching PubMed, web of science, China National Knowledge Infrastructure (CNKI) databases, a total of 11 articles including 13 independent studies were selected. The pooled odd ratios (ORs) along with their corresponding 95% confidence interval (CI) were estimated for allelic comparisons, additive model (homozygote comparisons; heterozygote comparisons), dominant model and recessive model. We also assessed the heterogeneity across the studies and publication bias. RESULTS: The results of combined analysis revealed a significantly increased risk of TB for single nucleotide polymorphism (SNP) rs9061 in all five comparisons (allelic comparisons: OR=1.28, 95% CI=1.14–1.44, p<0.0001; homozygote comparisons: OR=2.84, 95% CI=1.84–4.38, p<0.00001; heterozygote comparisons: OR=1.23, 95% CI=1.05–1.43, p=0.009; dominant model: OR=1.32, 95% CI=1.14–1.53, p=0.0003; recessive model: OR=2.26, 95% CI=1.18–4.34, p=0.01). In subgroup analysis, the risk of TB associated with SNP rs9061 appeared to be increased. Moreover, increased risk of TB was also found in Asian subgroup of SNP rs11556887, while decreased risk of TB appeared in large sample size subgroup of SNP rs1135791. No significant association was observed between other SNPs and the risk of TB. CONCLUSION: Our meta-analysis suggested that the variant of SNP rs9061 might be a risk factor for TB.

ABSTRACT

Objective To investigate the impact of CYP3A5 and ABCB1 polymorphisms on the initial individualized treatment with tacrolimus (FK506) in renal transplant recipients during switching from cyclosporine (CsA) to FK506. Methods Polymerase chain reaction and restriction fragment length polymorphism method (PCR-RFLP) was employed to investigate CYP3A5 (A6989G) and ABCB1 (exon12 [C1236T], exon21G[A] 2677T and exon26 [C3435T]) genotype data. The initial trough concentration/ dose (Co/D) values were compared among different CYP3A5 genotypes in renal transplant recipients switching from CsA to FK506 using one-way ANOVA. In addition, the initial C0 /D values were also compared among different ABCB1 (exon12[C1236T], exon21G[A]2677T and exon26[C3435T]) genotypes and their haplotypes using one-way ANOVA. Results The FK506 C0 /D values were significantly different between different CYP3A5 genotypes (AA, AG and GG) at the 7th, 14th, 21th and 28th day of conversion from CsA to FK506 in renal transplant recipients CP

ABSTRACT

<p><b>OBJECTIVE</b>To identify the uptake and biological distribution of technetium galactosyl human serum albumin diethylenetriamine pentaacetic acid injection (99mTc-GSA) in three mouse models with different degrees of hepatic injuries.</p><p><b>METHODS</b>Three mouse models including hepatic fibrosis, hepatic cholestasis, and liver cancer were established. Hepatic fibrosis model was established by intraperitoneal injection of carbon tetrachloride, 0.4 ml 10%, every 48 hours for 48 days. Hepatic cholestasis model was set up by ligature of the common bile duct for 72 hours, and liver cancer model by implantation of H22 tumor cells underneath liver capsule for 10 days. On measurement, each mouse in different models and normal controls was injected with 0.1 ml (0.37 MBq)99mTc-GSA (2 microg) into vena caudalis, and 5 minutes later sacrificed by decapitation. Important organs and tissues including liver, heart, lungs, kidney, spleen, stomach, blood, bones, muscles, and intestines were taken and their different radio countings were measured. The hepatic injuries were evaluated with serum and pathological examinations.</p><p><b>RESULTS</b>99mTc-GSA was concentrated in the liver in all three models and the control mice ( >40% ID x g(-1)). Compared with the control mice (90.05 +/- 10.55)% ID x g(-1), the density of 99mTc-GSA was significantly lower in the models with hepatic injuries (P < 0.001). The liver function test indicated that the injury in hepatic fibrosis model was less serious than those in the other two models. However, the concentration of 99mTc-GSA in hepatic fibrosis model [(72.20 +/- 2.13)% ID x g(-1)] was significantly higher than those in the models with cholestasis [(56.72 +/- 5.92)% ID x g(-1)] and liver cancer [(42.80 +/- 6.05)% ID x g(-1)] (P < 0.001).</p><p><b>CONCLUSIONS</b>99mTc-GSA may well concentrate in liver and its concentration degree is adversely correlated with hepatic injuries. Therefore 99mTc-GSA may be clinically used as liver imaging agent. When combined with three-dimensional scanning technique, it may facilitate constructing a new three-dimensional imaging method to demonstrate the function of designed liver segments.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the safety, biodistribution, and dosimetry of myocardial perfusion imaging agent 99Tc(m)N-NOET in 10 healthy volunteers.</p><p><b>METHODS</b>744-792 MBq of 99Tc(m)N-NOET was injected to each volunteer. Safety parameters and adverse event was measured in 24 hours of injection. Biodistribution was studied by whole-body imaging 1, 30 minutes, 1, 2, 4, 8, and 24 hours after the injection of 99Tc(m)N-NOET. The estimation of dosimetry was based on the standard medical internal radiation dose method using MIRDOSE 3.0 analysis program. Myocardial single photon emission computed tomography (SPECT) imaging was performed at 1 and 4 hours after injection.</p><p><b>RESULTS</b>No undesirable effects were reported by the subject during 24 hours after injection of 99Tc(m)N-NOET. No clinically significant changes were found in vital signs (heart rate, blood pressure, and electrocardiogram). No biochemical aspects and serology changes were measured. The myocardial SPECT imaging was clear. Cardiac uptake of 99Tc(m)N-NOET was as high as 2.68% at 2 hours after injection. The heart to lung ratio was more than 1 from 30 minutes after injection, reaching a maximum of 1.91 +/- 0.53 at 2 hours after injection. Radiation dosimetry calculations indicated an effective absorbed dose of 1.28 x 10(-5) Sv/MBq. The dosimetry in each main organ is lower then 50 mGy given 740 MBq of 99Tc(m)N-NOET in once imaging.</p><p><b>CONCLUSIONS</b>99Tc(m)N-NOET exhibits high cardiac uptake and low estimated effective absorbed dose. It's a safe myocardial perfusion imaging agent.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To assess the value of (99m)Tc-N-NOET ((99m)Tc-N-ethoxy-N-ethyl dithiocarbamato-nitrito) myocardial perfusion SPECT for the diagnosis of coronary artery disease.</p><p><b>METHODS</b>A total of 42 patients [mean age (54 +/- 9) years, 35 men] with suspected chest pain were included in this study. 740 MBq of (99m)Tc-N-NOET was injected intravenously during bicycle exercise when the heart rate attained reached more than 85% of the expected maximum, or in cases of angina pectoris, severe arrhythmias and ischemic ST segment changes. (99m)Tc-N-NOET 740 MBq, SPECT myocardial imaging acquisitions were obtained at 15 minutes and 2 hours after (99m)Tc-N-NOET injection. Coronary angiography was performed in all patients.</p><p><b>RESULTS</b>Coronary artery stenosis was detected in 26 patients and normal coronary angiography was shown in 16 patients. (99m)Tc-N-NOET myocardial perfusion imaging was abnormal in twenty-one patients out of the 26 patients with significant coronary artery stenosis (sensitivity, 81%); 14 out of 16 patients with normal angiography had a normal myocardial perfusion imaging (specificity, 88%). The positive predictive value, negative predictive value and predictive accuracy of (99m)Tc-N-NOET myocardial perfusion imaging for detection of CAD was 91%, 74% and 83%, respectively. The sensitivity of the imaging for detecting single vessel, double vessels and triple vessels disease were 60% (6/10), 86% (6/7) and 100% (9/9), respectively. There was mild (99m)Tc-N-NOET lung uptake in patients with coronary artery stenosis 15 minutes post (99m)Tc-N-NOET injection.</p><p><b>CONCLUSION</b>SPECT myocardial perfusion imaging with (99m)Tc-N-NOET supplied an important diagnostic tool for detecting coronary artery disease. Lung uptake with stress (99m)Tc-N-NOET might be related to coronary artery disease.</p>

ABSTRACT

<p><b>OBJECTIVE</b>(99m)Tc-[bis (dimethoxypropylphosphinoethyl)-ethoxyethylamine (PNP5)]-[bis (N-ethoxyethyl)-dithiocarbamate (DBODC)] nitride ([(99m)Tc (N) (PNP5) (DBODC)](+)) is a new myocardial perfusion tracer with high heart uptake and rapid liver clearance. The objectives of this study were to compare the myocardial imaging of [(99m)Tc (N) (PNP5) (DBODC)](+) with (99m)Tc-MIBI in a canine model of acute myocardial ischemia.</p><p><b>METHODS</b>The left anterior descending artery (LAD) was occluded in 12 adult beagle dogs, adenosine was then infused intravenously at a rate of 0.14 mg.kg(-1).min(-1) for 6 min. At the end of 3 min of adenosine infusion, 185 MBq of [(99m)Tc (N) (PNP5) (DBODC)](+) or (99m)Tc-MIBI was injected intravenously. The occluder was released after 6 min adenosine infusion. Serial myocardial SPECT imaging acquisitions were obtained at 0.5, 1, 1.5 and 2 h after tracer injection, respectively. Rest myocardial SPECT imaging was acquired in the next day.</p><p><b>RESULTS</b>Similar as (99m)Tc-MIBI, [(99m)Tc (N) (PNP5) (DBODC)](+) exhibited high heart uptake, minimal lung uptake and minimal redistribution. No significant myocardial washout was observed with both tracers over a period of 2 hours. [(99m)Tc (N) (PNP5) (DBODC)](+) clearance from the liver was more rapid than that with (99m)Tc-MIBI (heart-liver radio at 60 min, 1.36 +/- 0.43 vs. 0.58 +/- 0.21, P = 0.005). [(99m)Tc (N) (PNP5) (DBODC)](+) the ability to detect myocardial ischemia was also comparable between the two tracers ([(99m)Tc (N) (PNP5) (DBODC)](+) detected 3.60 +/- 1.52 defect segments, (99m)Tc-MIBI detected 4.25 +/- 0.96 defect segments, P = 0.48). The image quality of [(99m)Tc (N) (PNP5) (DBODC)](+) was better than (99m)Tc-MIBI.</p><p><b>CONCLUSION</b>[(99m)Tc (N) (PNP5) (DBODC)](+) is comparable to (99m)Tc-MIBI on detecting myocardial ischemia in this model and liver clearance is more rapid and the image quality is better than (99m)Tc-MIBI.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of Shenkang Injection (SI) on hypertrophy and expressions of p21 and p27 in glomerular mesangial cells of rats cultured in high glucose fluid.</p><p><b>METHODS</b>Cultured rat glomerular mesangial cells were divided into 6 groups: the normal control group, mannitol-treated group, high glucose-treated group, high, middle and low dose SI treated groups. The synthesis of cell protein and DNA were determined with incorporation of 3H-TdR and 3H-Leu. The changes of p21 mRNA and expressions of p21 and p27 protein were determined by reverse transcriptase polymerase chain reaction and Western blot.</p><p><b>RESULTS</b>High glucose caused incorporation of 3H-TdR reduced, incorporation of 3H-Leu increased. SI could decrease incorporation of 3H-Leu, increase incorporation of 3H-TdR. Compared with the high glucose-treated group, SI could inhibit the overexpression of p2l mRNA and protein,p27 protein induced by high glucose.</p><p><b>CONCLUSION</b>SI could inhibit hypertrophy of glomerular mesangial cells cultured in high glucose partly through inhibiting overexpressions of p2l mRNA, p21 and p27 protein.</p>

ABSTRACT

To construct gene vaccine of PPV and to investigate the effects of interleukin 2 (IL-2) as an adjuvant on immune responses in mouse, the recombinant expression plasmid of pCIneo-IL2-VP2 was constructed and transfected into PK-15 cells by lipofectamine, the expressed product was detected by immunofluore assay. To study the immune effects of DNA vaccine in vitro and in vivo, mice were used as the animal model. The recombinant plasmid pCIneo-IL2-VP2, the control plasmid pCI-neo and the PPV live vaccine were immunized by intramuscular injection. Anti-PPV antibodies were measured by ELISA, lymphocyte proliferation activity was detected using MTT method, and the specific killing activities of CTL were assayed too. The results show that the immunized mice produced PPV antibody after one week, and reached to highest after four weeks. Compared with the control group, the pCIneo-IL2-VP2 immunized group produced significant differences in the antibody titers, the lymphocyte proliferation activity and the specific killing activities of CTL. The pCIneo-IL2-VP2 induced humoral and cellular immunity responses similarly to that the live vaccine induced. These results manifested that the PPV DNA vaccine successfully induced humoral and cellular immunity response in mice with the IL-2 gene as an adjuvant.
